Latest Retatrutide Research
A dated record of what has actually been reported, labelled by evidence tier — and what is still to come.
How entries are labelled
Peer-reviewed published in a journal after independent review. Sponsor announcement released by the developer before full publication — useful, but preliminary. Pending expected but not yet reported; we publish no figures for these.

TRIUMPH-1 published in the New England Journal of Medicine
Peer-reviewed “Retatrutide, a Triple Hormone Receptor Agonist, for Treatment of Obesity”, Jastreboff AM et al., DOI 10.1056/NEJMoa2604169. The abstract reports a 25.0% mean weight reduction at 12 mg against 3.9% for placebo — the treatment-regimen estimand, which is why it differs from the 28.3% figure in the May announcement. This is the first retatrutide Phase 3 trial to undergo independent peer review.

TRIUMPH-2 and TRIUMPH-3 topline; US filing planned for Q1 2027
Sponsor announcement In adults with type 2 diabetes, weight reduction reached 20.8% at 80 weeks with A1C reductions up to 1.6%. In adults with severe obesity and established cardiovascular disease, weight reduction reached 22.6%. Cardiovascular event analyses were reported with confidence intervals crossing 1.0 and small event counts; they establish neither benefit nor harm. Eli Lilly said it was completing the manufacturing data package and planned to submit a Biologics License Application to the US FDA in Q1 2027.

TRIUMPH-1 topline results
Sponsor announcement All three doses met the primary and key secondary endpoints. Mean weight reduction at 80 weeks was 19.0%, 25.9% and 28.3% for 4 mg, 9 mg and 12 mg against 2.2% for placebo (efficacy estimand). In a pre-specified extension of 532 participants with baseline BMI ≥35, reduction reached 30.3% at 104 weeks. Discontinuation due to adverse events was 4.1% at 4 mg — below the 4.9% placebo rate.

TRIUMPH-4 — the programme's first Phase 3 readout
Sponsor announcement In 445 adults with obesity and knee osteoarthritis, mean weight reduction at 68 weeks was 26.4% and 28.7% for 9 mg and 12 mg against 2.1% for placebo, with WOMAC pain subscale reductions of 4.5 and 4.4 points against 2.4. This trial also reported the programme's highest dysesthesia rate, 20.9% at 12 mg against 0.7% on placebo.

Phase 2 obesity results published in the NEJM
Peer-reviewed A 48-week trial in 338 US adults. Mean weight reduction was 17.5% at the 24-week primary endpoint and 24.2% at 48 weeks. The lead investigator noted participants “had not yet reached a weight plateau” when the trial ended — the observation that shaped the longer Phase 3 designs.

First-in-human study completed
Registry record NCT03841630, a single-ascending-dose safety, tolerability and pharmacokinetics study in 45 healthy participants, completed July 2019. The start of the clinical programme.

Recorded dates are not promises
Primary completion dates come from the trial registry and change. Completion is also not the same as reporting: topline announcements typically follow months later, and peer-reviewed publication later still. Likewise, Eli Lilly's stated plan to file in Q1 2027 is a company plan, not a regulatory event — and filing is not approval.
How this page is maintained
- Check the ClinicalTrials.gov register for status and last-update changes across all retatrutide records.
- Check Eli Lilly's investor newsroom for new announcements.
- Check for peer-reviewed publication of any trial previously reported only as topline.
- Record, for each new item: date, source, study identifier, phase, population, design, duration, endpoints, findings as stated, safety observations, limitations, and whether it is peer-reviewed or preliminary.
- Update the verification date only after sources have actually been rechecked.
